Sample Requirements and Shipping Information

Sample Requirements

 

Please label every sample with the patient’s first and last name and date of birth.

  • For all molecular genetic analyses (single genes and multi-gene panels):
    • EDTA blood or DNA or prenatal samples
  • For chromosomal microarray (CMA):
    • EDTA blood or DNA or prenatal samples or products of conception
  • For karyotyping:
    • heparinized blood or skin biopsy or prenatal samples
  • For fluorescence in situ hybridization (FISH): 
    • heparinized blood
  • For microsatellite instability (MSI):
    • tumor tissue

DNA*:

  • Preferred quantity: 10 µg (minimum required: 3 µg)
  • Packaging: it is best to use a transport tube with a screw cap; please use Parafilm to ensure the tube is tightly sealed
  • Shipping temperature: room temperature or refrigerated (not frozen!)
  • Transport time: your sample should arrive at MGZ within 4-5 days
  • *as part of the pre-analysis, DNA concentration and quality are measured; if the sample is insufficient, the analysis cannot be performed

EDTA blood:

  • Preferred quantity: 4 ml (minimum required: 2 ml)
  • Packaging: shatter-proof; please use monovettes (Sarstedt) or vacutainers (Becton-Dickinson), preferably without a separating gel
  • Shipping temperature: room temperature or refrigerated (not frozen!)
  • Transport time: your sample should arrive at MGZ within 4-5 days

Heparinized blood (either lithium or sodium heparin):

  • Preferred quantity: 4-5 ml (minimum required: 1-2 ml)
  • Packaging: shatter-proof; please use monovettes (Sarstedt) or vacutainers (Becton-Dickinson), preferably without a separating gel
  • Shipping temperature: room temperature
  • Transport time: your sample should arrive at MGZ within 2-4 days

Prenatal samples^:

Chorionic villi sample (CVS)^:

  • For molecular genetic testing:
    • Either >10 mg or >3 chorionic villi in a tube with sterile transport media,
    • or 1-2 sterile confluent T25 flask(s) of cultured CVS cells
  • For cytogenetic testing:
    • Either >15 mg or >5 chorionic villi in a tube with sterile transport media,
    • or 1-2 sterile confluent T25 flask(s) of cultured CVS cells
  • Packaging: shatter-proof
  • Shipping temperature: room temperature or refrigerated (not frozen!)
  • Transport time: your sample should arrive at MGZ within 2-3 days

Amniotic fluid^:

  • For molecular genetic testing:
    • Either preferred quantity: 10 ml (minimum required: 5 ml),
    • or 1-2 sterile confluent T25 flask(s) of cultured amniotic fluid cells
  • For cytogenetic testing:
    • Either preferred quantity: 10-20 ml (minimum required: 5 ml),
    • or 1-2 sterile confluent T25 flask(s) of cultured amniotic fluid cells
  • Packaging: shatter-proof
  • Shipping temperature: room temperature or refrigerated (not frozen!)
  • Transport time: your sample should arrive at MGZ within 2-3 days

Fetal cord blood^:

  • Preferred quantity: >1 ml (minimum required: 0.5 ml) heparinized blood (either lithium or sodium heparin)
  • Packaging: shatter-proof; please use monovettes (Sarstedt) or vacutainers (Becton-Dickinson), preferably without a separating gel
  • Shipping temperature: room temperature or refrigerated (not frozen!)
  • Transport time: your sample should arrive at MGZ within 2-3 days
  • ^a maternal EDTA blood sample must accompany all prenatal samples for maternal contamination studies; see above for EDTA blood sample requirements

Products of conception:

  • Preferred quantity: either >30 mg or >10 chorionic villi in a tube with sterile transport media; or fetal tissue
  • Packaging: shatter-proof
  • Shipping temperature: room temperature or refrigerated
  • Transport time: your sample should arrive at MGZ within 2-3 days

Skin biopsy:

  • Preferred quantity: ~2 mm x 2mm to 3 mm x 3 mm biopsy
  • Packaging: shatter-proof
  • Shipping temperature: room temperature
  • Transport time: your sample should arrive at MGZ within 2-4 days

Muscle biopsy*:

  • Preferred quantity: ~2 mm x 2mm to 3 mm x 3 mm muscle needle biopsy
  • Packaging: ~25 cm x 25 cm Styrofoam box (filled with dry ice)
  • Shipping temperature: we recommend you send muscle biopsies on dry ice with an express courier
  • Transport time: your sample should be delivered to MGZ within 2-3 days; take care to send the package on a Monday or Tuesday so it does not end up sitting at the post office thawing over a weekend.
  • *please notify us you are sending a muscle biopsy at least one day in advance by emailing info@mgz-muenchen.com.

Tumor tissue*:

  • Sample types: we accept FFPE slides or blocks, and fresh tumor tissue
  • Packaging:
    • FFPE: not critical
    • Fresh tissue: shatter-proof; in a suitable buffer solution
  • Transport time:
    • FFPE: not critical
    • Fresh tissue: your sample should be delivered to MGZ within 1-2 weeks
  • Shipping temperature: room temperature
  • *if you are interested in sending a tumor sample for MSI, please first email inquiry@mgz-muenchen.com to discuss the case

To ensure prompt analysis of your sample, in your package please make sure to include a completed Test Request Form and a cost statement.

Samples may be sent by post or courier and may be delivered between 8 AM and 5 PM, Monday through Friday.

On the customs declaration form, note that you are sending a specimen that is not infectious, not poisonous, not radioactive, not toxic. The monetary value of the specimen may be written as 1€.

Before sending prenatal or other urgent samples, we ask that you notify us be emailing inquiry@mgz-muenchen.com.

Once we have received your shipment, you will receive an email confirmation of sample receipt.

Insufficient Samples

 

In some cases, samples cannot be analyzed or will be analyzed only with reservations.

When can a sample not be analyzed?

  • The patient's identity cannot be discerned (unlabeled samples, no Test Request Form)
  • Badly damaged sample container (e.g. leakage)
  • Obviously non-sterile or contaminated sample
  • Wrong material

When may a sample be analyzed only with reservations?

  • Test request accompanied by an unlabeled or unlcearly labeled sample
  • Too little material
Remaining, Unused DNA

 

After the completion of molecular genetic analyses, all remaining unused DNA will be stored on location and is thus available for future testing. If the remaining unused DNA should be destroyed after testing, please indicate this clearly on the Test Request Form.
